It is true that the hair follicle has a “biological clock.” But that does not mean the follicle works more at night or that hair-care products are automatically more effective when applied at night.

In a 2014 human hair-follicle study, experimental reduction of BMAL1 or PER1 activity prolonged anagen. This suggests that circadian-clock proteins can modulate the human hair cycle. The study did not, however, demonstrate that human hair follicles are more active at night.
What Did the 2013 PNAS Study Actually Show?
The frequently cited 2013 PNAS study on circadian rhythm and hair follicles was conducted mainly in mice. Researchers identified peripheral clock activity and a pronounced daily rhythm of mitosis in the matrix and dermal-papilla regions of anagen follicles.
The crucial detail is that, in this experimental model, mitotic activity and hair-growth rate were higher in the morning and lower in the evening. The study therefore does not support the claim that the follicle’s most active growth period is at night. Instead, it shows that follicular biology can be time-regulated and that the timing depends on species and experimental conditions.
Clock-time findings from mice should not be mapped directly onto the human scalp as equivalent hours of the day.
Does the Follicle Work at Night or During the Day?
Short answer: both. The hair follicle is a living mini-organ that maintains metabolic and signaling activity throughout the 24-hour day. Some cellular processes may fluctuate in a circadian manner, but current evidence does not justify describing the human hair follicle as an organ that “works at night.”
Statements such as “cell division speeds up at night,” “night is the follicle’s most active renewal period,” or “the follicle works less during the day” are therefore too definitive.
If Growth Hormone Rises During Sleep, Do Peptides Work Better at Night?
Sleep-associated pulses of growth hormone are part of normal human physiology. But this does not establish that cosmetic peptides applied to the scalp, such as Biotinoyl Tripeptide-1 or Acetyl Tetrapeptide-3, work synergistically with nighttime growth-hormone peaks.

Is DNA Repair More Active at Night?

Melatonin and the Hair Follicle
The relationship between melatonin and hair biology has been studied. In a small double-blind, placebo-controlled pilot study published in 2004, 40 women used either 0.1% topical melatonin or placebo once daily for six months. Significant increases in anagen-hair rate were reported in some subgroups.
This result is interesting, but three limitations matter:
- The study was a small pilot trial.
- It specifically investigated topical melatonin.
- It did not test whether nighttime application was superior to daytime application.
